The strength trial everyone quotes: 13 men, 6 days, 600 mg

In a double-blind, placebo-controlled crossover (a human RCT, top of the evidence ladder, bottom of the size ladder), 13 college-aged men took 600 mg of alpha-GPC daily for 6 days, and their lower-body isometric mid-thigh pull force improved versus placebo: a gain of 98.8 newtons against a 39.0 newton decline on placebo, p = 0.044. The upper-body test moved in the same direction without reaching significance (p = 0.127). That result is real, and it is also 13 people pulling on an immovable bar after six days of capsules: a promising pilot, not a proven effect.

The bigger trial that found less

Two years later, 48 healthy men were randomized to 7 days of alpha-GPC 500 mg, alpha-GPC 250 mg, caffeine 200 mg, or placebo, and the result sheet reads differently: no differences in isometric strength or psychomotor vigilance on any arm. The 250 mg group did improve countermovement-jump velocity and power, the 500 mg group did not, and the 500 mg group showed significantly lowered serum thyroid-stimulating hormone (p < 0.04). Read that pattern honestly: the lower dose moved one jump metric, the higher dose moved a thyroid marker instead of a muscle. Small studies often look exactly like this.

The only trial in women is also the outlier in design: a randomized crossover in 12 overweight or obese women found a single 1000 mg dose taken before sprint-interval exercise sped up heart-rate-variability and blood-pressure recovery afterwards (human RCT). The blend problem: two trials cannot credit alpha-GPC at all

The remaining two trials tested cocktails. In one, 20 trained college-aged men took a single dose of a blend carrying alpha-GPC 500 mg alongside uridine monophosphate and DHA, 90 minutes before testing: no improvement in vertical jump, bench-press repetitions, or cognition versus placebo (Bunn 2018, human RCT, crossover). A clean null.

In the other, 30 trained male cyclists took 7 days of BCAAs 8 g plus L-citrulline 6 g plus alpha-GPC 300 mg, and their 20-km time-trial peak power rose (354 vs 322 watts, p = .003) while time to fatigue stretched from about 14.5 to almost 20 minutes (p = .001), with completion time and average power unchanged (Harrington 2023, human RCT, crossover). In 1992, researchers gave young and elderly volunteers GHRH, the releasing hormone that triggers growth hormone, with or without alpha-GPC, and alpha-GPC potentiated the growth hormone response in both groups, more strongly in the elderly (Ceda 1992). This was a non-randomized acute hormone-challenge study: a physiology probe, not an outcome trial.

In 2012, a double-blind randomized crossover in 8 healthy young men found a single 1000 mg dose of GPC raised plasma growth hormone at the 60-minute mark and raised free fatty acids and ketone bodies, markers of fat oxidation, at 120 minutes, while placebo changed nothing (Kawamura 2012).

Now the part the forum posts skip: a transient growth hormone pulse is not a muscle-gain result. Growth hormone also spikes when you sleep, sprint, or skip breakfast.
