Why there is no official alpha-GPC dose

In the United States, alpha-GPC is sold as a dietary supplement, and supplements do not carry an official dose the way prescription products do. No recommended intake has ever been set for alpha-GPC itself. Every number you have seen on a label, a forum, or a dosage calculator is somebody's choice, not an authority's.

The deeper reason dosage pages disagree is that the research splits into two literatures that used very different amounts. The clinical literature comes from countries where alpha-GPC has a prescription history, and it dosed high and long: 1,200 mg per day, for months, in older patients under medical supervision. The modern healthy-adult literature dosed lower and shorter: single doses and courses of a week or two, mostly between 250 and 1,000 mg. What the studies actually used, dose by dose

The flagship is the trial nearly every dosage page quotes without citing: a 180-day double-blind randomized trial in 261 patients with mild to moderate Alzheimer's dementia, which used 1,200 mg per day and reported improved ADAS-Cog scores while the placebo group worsened (PMID 12637119).

Three things the table alone would hide. A further open-label series in the same memory-clinic population used the standard 1,200 mg per day without reporting a duration (PMID 28745666). Several mid-tier studies used 800 mg per day, including a 6-month double-blind randomized trial in 49 older adults with type 2 diabetes (PMID 40095622) and an 11-month observational study in 34 hearing-aid users (PMID 34149400). And two athletic results came from blends: alpha-GPC 500 mg with uridine monophosphate and DHA dosed 90 minutes before testing (PMID 28222577), and 300 mg inside a BCAA and citrulline stack over 7 days (PMID 37229544), so neither result can be attributed to alpha-GPC alone.

One boundary matters most: the dementia-comparison and stroke-rehabilitation literature also contains 1,000 mg per day given by intramuscular or intravenous route in clinical settings. Those are medical protocols, not supplement servings, and they do not translate to capsules.

Capsules on the US market cluster around 300 and 600 mg, so this is the comparison people actually shop with. The two sizes map to the evidence differently.

600 mg per day is the only dose tested in both a modern cognition RCT and a strength RCT. The 2024 Korean trial randomized 100 adults with amnestic mild cognitive impairment to 600 mg per day of soy-derived alpha-GPC or placebo for 12 weeks and reported significantly better ADAS-cog scores with no difference in adverse-event rates (PMID 39300341). On the athletic side, a 6-day double-blind crossover in 13 college-aged men used the same 600 mg per day and measured higher lower-body isometric peak force versus placebo, with the upper-body test trending better without reaching significance (PMID 26582972). Both studies are small, but the dose is at least directly tested.

Exactly 300 mg has never been tested on its own in the 144-study library. The nearest data points are cousins: a single 315 mg dose improved Stroop scores 60 minutes after ingestion in 20 resistance-trained men (PMID 39683633), and 250 mg per day for 7 days improved countermovement-jump velocity and power in a 48-man randomized trial (PMID 29042830). Those are acute or one-week results in healthy adults, not evidence about months of use.

Note what neither capsule reaches: the 1,200 mg per day the prescription-era clinical trials used. A 600 mg serving is half that daily figure, and a 300 mg serving is a quarter of it. That fact makes no capsule right or wrong; it tells you which literature a label is echoing.

Timing: what the trials actually did

The one-hour pattern in acute studies

When researchers dosed once and measured, the measurements clustered about an hour later. The acute cognition crossover recorded Stroop improvements 60 minutes after ingestion of 630 mg or 315 mg (PMID 39683633). The growth-hormone study in 8 young men found plasma growth hormone elevated at 60 minutes and fat-oxidation markers raised at 120 minutes after a single 1,000 mg dose (PMID 22673596). A crossover in 12 women used a single 1,000 mg dose before sprint interval exercise and reported faster recovery of heart-rate variability and blood pressure afterwards (PMID 36235623).

The one pre-exercise protocol with an exact clock time is a negative result: a blend carrying alpha-GPC 500 mg, dosed 90 minutes before testing, improved nothing versus placebo in 20 trained men (PMID 28222577). And the only minute-by-minute absorption anchor is intramuscular, not oral: free plasma choline peaked 15 to 30 minutes after a 1,000 mg intramuscular dose (PMID 1428296), which says nothing about capsules. The strength RCT dosed 600 mg per day for 6 days and then tested. The dose-comparison trial ran 250 or 500 mg per day for 7 days. The clinical trials ran 600 to 1,200 mg per day for 12 weeks to 24 months, with the prescription-era protocols reaching their 1,200 mg total in divided doses across the day rather than in one serving. None of the study records specifies a clock time.

Food, morning, evening: honestly unanswered

No study record in the library reports whether oral doses went in with food or without, and none compares morning against evening dosing. That is the honest end of the timing evidence. When a page hands you a food rule or a clock rule for alpha-GPC, it is stating a preference, not a study result.

What US labels typically deliver

On the US market, standalone alpha-GPC capsules commonly deliver 300 to 600 mg per serving. That is a market observation, not a recommendation, and this section stays descriptive on purpose: MAXXING sells no alpha-GPC product, so we have no serving size to defend.

How much is too much? What the data can and cannot say

The highest daily oral dose in the library is 1,200 mg, used in supervised clinical trials for up to 24 months (PMID 35719710). The highest single oral doses are the 1,000 mg used in the acute studies above.

EFSA's 2026 novel-food opinion assessed soy-derived alpha-GPC at intakes up to 203.7 mg per day and concluded it does not raise safety concerns at that level for people over 3 years of age, working from a rat no-observed-adverse-effect level of 1000 mg per kilogram per day and noting limitations in the animal data (PMID 42131868). Read the gap plainly: the assessed supplement intake is lower than typical US servings. That is a statement about what has been evaluated, not a danger claim in either direction.

Two dose-linked human signals belong on this page. The biggest gap is the one this page cannot fill: there is no long-term dosing evidence in healthy adults. The longest single-ingredient alpha-GPC trial in healthy people in this library ran two weeks, a 39-person motivation study (PMID 34207484); before that sits a 10-day study from 1991 in 32 young volunteers (PMID 2071257). Nobody has tested what months of 300, 600, or 1,200 mg per day does in a healthy 30-year-old. At any dose. Ever.
